Principle: The method is based on the observation that Lactobacillus plantarum requires nicotinic acid (or nicotinamide) for growth. Using a basal medium complete in all respects except for nicotinic acid, growth responses of the organism are compared quantitatively in standard and unknown solutions. The acid produced by the organism is measured to determine the extent of growth and thereby the amount of vitamin in the test solution.
